How to Inoculate a Sterilized Grain Bag

Growing mushrooms successfully starts with proper inoculation. By following sterile technique and carefully preparing your work area, you can greatly reduce the risk of contamination and ensure healthy colonization of your grain bag.


Step 1: Prepare Your Workspace

Before handling your grain bag, set up a clean working environment.

  • Work in a draft-free room.

  • Wipe down surfaces with isopropyl alcohol (70%). ⚠️ Safety Note: Never use alcohol in a closed space when working with an open flame. Keep alcohol containers away from heat sources.

  • Consider wearing a mask to reduce airborne contaminants.

  • Keep all necessary items (syringe, alcohol wipes, lighter) within reach.


Step 2: Open the Bag Carefully

  • The sterilized grain bag arrives sealed inside a protective outer poly bag.

  • Gently remove it from the outer packaging.

  • Do not open the grain bag itself—keep the sterile seal intact until inoculation.


Step 3: Put on Gloves and Sanitize the Injection Port

  • Wash your hands thoroughly with soap and warm water.

  • Put on clean, disposable gloves.

  • Do not peel off or puncture the clear plastic film covering the self-healing injection port.

  • Use an alcohol wipe to clean the injection port thoroughly, then allow it to air-dry for a few seconds. ⚠️ Safety Note: Alcohol is flammable. Allow it to fully evaporate before exposing the port to a flame.


Step 4: Prepare Your Syringe

  • Shake the syringe gently to distribute spores or liquid culture evenly.

  • Remove the protective cap from the syringe needle without touching the needle itself.

  • If the needle is sterile straight from the package, you may use it for a one-time injection without flame sterilizing.

  • If you plan to reuse the needle or syringe for multiple bags, flame sterilize the needle until it glows red using a lighter or alcohol lamp. ⚠️ Safety Note: Never flame sterilize near alcohol containers or freshly wiped surfaces. Always work in a well-ventilated space.

  • Allow the needle to cool for 5–10 seconds before injecting.


Step 5: Inoculate the Grain Bag

  • Insert the sterilized (or fresh sterile) needle straight through the injection port.

  • Slowly inject 2–5 mL of liquid culture or spore solution into the grain.

  • Withdraw the needle carefully and recap it.

  • Wipe the injection port again with alcohol to maintain sterility. ⚠️ Safety Note: Ensure the port is dry before introducing any flame near your work area.


Step 6: Mix and Store

  • Gently massage the bag to distribute the solution into the grains, but avoid breaking up kernels too much.

  • Seal the outer poly bag again or place the grain bag in a clean tote for added protection.

  • Store the inoculated bag in a dark area at 70–78°F (21–26°C) for colonization.


Additional Tips for Success

  • If using multiple injections from the same syringe, always flame sterilize the needle between bags.

  • Do not over-shake the bag immediately after inoculation—allow the mycelium to establish before breaking it up later.

  • Watch for signs of healthy colonization: white, threadlike growth spreading from the inoculation points.

  • Discard bags showing green, black, or foul-smelling contamination.

Inject Culture
Insert your syringe through the injection port and inject the liquid directly into the middle of the bag.

  • Use 3–10 cc of liquid culture (recommended)

  • Use no more than 2-4 cc of spores (not preferred)

Do not shake the bag after injection.

Let It Grow
Keep the bag in a dark place at 70–75°F. Mycelium will begin to grow in a few days. Avoid moving or disturbing the bag.

Break & Shake
Once 20–30% of the grain is covered in white mycelium, gently break up and mix the contents by massaging the bag from the outside.

Spawn to Substrate
Once fully colonized, mix the grain with a bulk substrate like CVG in a clean container. Allow it to colonize again before fruiting.
